Antioxidant Activities of Plant Extracts (Ammannia multiflora, Ammannia coccinea, and Salix gracilistyla) Activate the Nrf2/HO-1 Signaling Pathway

To identify potent plant extracts with strong antioxidant activity, we evaluated the free radical scavenging activity of 184 obtained from Freshwater Bioresources Culture Collection (FBCC) Nakdonggang National Institute Biological Resources (Republic Korea), as various have been used therapeutically to prevent chronic diseases associated oxidative stress. From them, three (FBCC-EP858 Ammannia multiflora, FBCC-EP920 coccinea, and FBCC-EP1014 Salix gracilistyla) were selected based on their abilities scavenge 2,2-diphenyl-1-picrylhydrazyl (DPPH) more than 80% efficiency. We found that these had in vitro half maximal inhibitory concentration (IC50) values ranging 11.89 14.26 μg/mL total (corresponding approximately 0.18, 0.22, 0.23 mM Trolox, respectively). also studied effect RAW 264.7 macrophages significantly downregulated relative cell viability at a 100 μg/mL. However, other two extracts, FBCC-EP858 FBCC-EP1014, did not affect same concentration. Additionally, all inhibited hydrogen peroxide (H2O2)-induced reactive oxygen species (ROS) production depolarization mitochondrial membrane potential macrophages. An additional experiment zebrafish larvae showed reduced 2′,7′-dichlorodihydrofluorescein diacetate (DCFDA) fluorescent intensity induced by H2O2. The upregulated expression nuclear factor erythroid 2-related 2 (Nrf2) heme oxygenase-1 (HO-1) expression, an HO-1 inhibitor, zinc protoporphyrin (ZnPP), attenuated extract-induced both vivo vitro. Taken together, findings suggest A. S. gracilistyla capacities activating Nrf2/HO-1 signaling pathway. These results could be useful for prevention treatment stress-mediated human diseases.
